Structural studies on Signal Transducing and Cytoskeletal Proteins

Abstract
The talk will present crystal structure of PH domain of non-receptor Bruton`s tyrosine kinase complex with IP4 and structure determination of double spectrin-like repeat from alpha-actinin. PH domain of Bruton`s tyrosine kinase targets the molecule to the membrane, where it binds to the polar head of PtdIns(3,4,5)P3, a costituent of the membrane. The crystal structure allows to understand the specificity of this PH domain towards the ligand, as well as gives a structural basis for understanding the role mutations involved in X-linked agammaglobulinemia - a severe immunodeficiency disease. The second part of the seminar will be focused on structure determination of a construct comprising two consecutive domains of a cytoskeletal protein: double spectrin-like repeat from alpha-actinin.
